Pantoprazole 40 mg + domperidone 10 mg
Pantoprazole is a drug acting as a proton pump inhibitor in the parietal cells of the stomach, which are in charge of acid secretion. It blocks the enzyme in the wall of the stomach that produces acid. By blocking the enzyme, the production of acid is decreased, and this allows the stomach and esophagus to heal. It is used in the treatment and prevention of acid peptic disease. Once ingested pantoprazole is activated in the gastric medium, and its absolute bioavailability is 77%. Pantoprazole diminishes the daily production of acid by 80- 95 %.
Domperidone
Domperidone is a potent dopamine receptor antagonist. It acts centrally and blocks the Dopamine receptor in the Chemoreceptor trigger zone and produces Antiemetic effect. It acts peripherally in the gastrointestinal system and increases oesophageal peristalsis, oesophageal sphincter pressure and gastric motility. These all facilitates gastric emptying. It is used in migraine to relieve nausea and vomiting.

- Gastroesophageal reflux disease
- Duodenal ulcer
- Gastric ulcer
- Dyspepsia
Dosage
1 tablet once daily.
Contraindications
- Hypersensitivity to pantoprazole or domperidone
- Patients with hepatic impairment
- Patients with gastrointestinal haemorrhage, obstruction
- In pregnant females
Precautions
- Tell the doctor if you are allergic to pantoprazole, dexlansoprazole (Dexilant), esomeprazole (Nexium), lansoprazole (Prevacid), omeprazole (Prilosec, Zegerid), rabeprazole (AcipHex), any other medications, or any of the ingredients in pantoprazole tablets
- Tell the doctor what prescription and nonprescription medications, vitamins, nutritional supplements, and herbal products you are taking or plan to take. Be sure to mention any of the following: certain antibiotics, including ampicillin (Principen), anticoagulants (blood thinners) such as warfarin (Coumadin), atazanavir (Reyataz), digoxin (Lanoxin, Lanoxicaps), diuretics (‘water pills’), iron supplements, ketoconazole (Nizoral), methotrexate (Rheumatrex, Trexall), and nelfinavir (Viracept).
- Tell the doctor if you have or have ever had a low level of magnesium in your blood.
- Tell the doctor if you are pregnant, plan to become pregnant, or are breast-feeding. If you become pregnant while taking pantoprazole, call your doctor.
- Not recommended in child12 yrs. Long-term therapy may lead to bacterial overgrowth in the GIT.
- Increases serum prolactin levels resulting to galactorrhoea in females and gynaecomastia in males. Hypertensive crisis in patients with phaeochromocytoma
Side effects
Headache, dizziness, nausea, vomiting, gas, joint pain, blistering or peeling skin, rash, hives, itching, swelling of the eyes, face, lips, mouth, throat, or tongue, difficulty breathing or swallowing, hoarseness, irregular, fast, or pounding heartbeat, excessive tiredness, dizziness, lightheadedness, muscle spasms, uncontrollable shaking of a part of the body, seizures, severe diarrhea with watery stools, stomach pain, fever
Drug interaction
Pantoprazole may reduce the absorption of ketoconazole, itraconazole, posaconazole, erlotinib when used concomitantly.
Co-administration of pantoprazole with atazanavir is not recommended.
Concurrent administration with MAO inhibitors is not recommended
